O67: PERFUSATE GLUCOSE REFLECTS TISSUE GLYCOGENATION DURING LIVER PERFUSION

Abstract Introduction Normothermic machine perfusion (NMP) is a method of organ preservation that aims to replicate the physiological environment, achieved by perfusing livers with blood-based perfusate at inflow pressures and temperature. NMP also permits viability assessment through evaluation flow rates portal vein hepatic artery. In addition this, biochemical gas analysis can be performed provide insights into metabolic activity liver. Method Discarded human liver grafts (n=6), were perfused for 24 hours. Core biopsies samples taken from each 5 distinct time intervals over fixed stained periodic acid-Schiff analysed Leica software quantitative estimate hepatocellular glycogen content. Result Hepatic concentration rose during first hour, followed steady decline thereafter until end perfusion. Contrary our initial hypothesis glucose within circuit would show an inverse relationship stores in cells, we found closely same trend. Conclusion Change hepatocyte content provides important insight synthetic function destined transplant. Our research suggests used as surrogate marker on valuable information glycogen-synthesising capability hepatocytes. future, this could potentially aid decision-making process regards graft transplant viability.
